weakness: This is correct. A WEAKNESS is a negative quality that is seen as undesirable. Example: My main WEAKNESS is my addiction to painkillers. "What is your greatest strength?" is used to ask candidates about positive/strong points; "What is your greatest weakness?" is used to ask candidates about negative/weak points.
lack: This is incorrect. A LACK is a deficiency. It is a synonym of WEAKNESS, but we tend to say A LACK OF something - A LACK OF money, A LACK of time - and don't use LACK on its own (what is your greatest LACK? = INCORRECT). Note: sentences like "intelligence is a LACK" or "this website has a LACK" are also incorrect. In these cases, we would probably use a verb (he LACKS intelligence, this website is LACKING something) or just use another term (this website has inadequacies or shortcomings).
failure: This is incorrect. A FAILURE is something that has not worked as hoped. Example: My poetry class for criminals was a complete FAILURE. FAILURE is the opposite of SUCCESS (not STRENGTH), therefore it is not the best answer here.
